The P2Y(14) receptor (P2Y(14)R) plays a key role in the modulation of inflammatory process, but very few classes of antagonists have been reported. A series of 3-amide benzoic acid derivatives were identified as novel and potent P2Y(14)R antagonists. The most potent antagonist, 16c, showed comparable activity (IC50 = 1.77 nM) to PPTN, the most potent P2Y(14)R antagonist reported. Compound 16c demonstrated dramatically improved aqueous solubility and excellent metabolic stability in rat and human microsomes. Investigation of the anti-inflammatory effect of 16c was performed in MSU treated THP-1 cells by flow cytometry, Western Blot and immunofluorescence labeling technology, which exhibited that 16c might be a promising candidate for further research. A set of new salts containing benzimidazole and pyrazine groups, namely, [HPZBI]X-+(-) nH(2)O (X = Cl (n = 1) (1), NO3 (n = 1) (2), ClO4 (n = 0) (3) and H2PO4 (n = 0) (4)) [PZBI = 2-(pyrazin-2-yl)-1H-benzimidazole], were obtained by the reaction of PZBI and a set of inorganic acid, respectively. The emission maxima can be found at 444, 472, 471, 432 and 443 nm for PZBI and its compounds 1-4 in the solid state at room temperature, respectively. Compared with the free PZBI, the emission maxima of compounds 1-4 are obviously blue/red-shifted for 1-4, indicating that the emission maxima are relative to the intermolecular packing distances. Their photoluminescent lifetime and quantum yield are 1.10 ns and 24.5% for 1, 0.99 ns and 26.4% for 2, 0.94 ns and 21.65% for 3, 2.86 ns and 20.57% for 4, respectively. Compounds 1-4 were structurally characterized by X-ray single crystal diffraction, FT-IR, and UV-Vis spectra. Single crystal X-ray diffraction reveals that pi center dot center dot center dot pi packing interactions and a set of hydrogen bonds can be observed. The shortest distances of pi center dot center dot center dot pi stacking interactions are 3.613, 3.534, 3.731 and 3.492 angstrom in compounds 1-4, respectively. Additionally, the thermal stabilities of compounds 1-4 were investigated in detail.

Pyrazinecarboxylic acid (PCA) was examined as a potential corrosion inhibitor for mild steel (MS) in 0.5 M HCl environment. The methods of electrochemical impedance spectroscopy (EIS), linear polarization resistance (LPR), as well potentiodynamic (PD) polarization were utilized. Furthermore, atomic force microscopy (AFM) and quantum chemical calculations were utilized. PD polarization curves demonstrated that PCA exhibited mixed inhibitor behavior. Scanning electron microscopy (SEM) offered the creation of an adsorptive layer on the surface of MS which prevented the steel against corrosive specimens. Furthermore, density functional theory (DFT) presented good agreement with electrochemical experimental results. The adsorption equilibrium constant (k(ads)) value was calculated to be 3.704 x 10(4) M-1 which was related to a high proportion of inhibitor on the surface. In the presence of 1.0 mM PCA, inhibition efficiency was determined as 95.2% from EIS results.

On the basis of crystal engineering, two similar esters of phenyl pyridine-2-carboxylate (I) and phenyl pyrazine-2-carboxylate (II) were designed and synthesized. The compounds were characterized using FT-IR, mass spectrometry, CHN-elemental analyses, NMR and PXRD. For each compound, two polymorphs were obtained (Ia, Ib and IIa, IIb) and identified by TGA, DSC and SCXRD. A comparison of the crystal structures of the polymorphs revealed that the different torsion angles of the pyrazine (tau(1)) and phenyl (tau(2)) rings to the ester backbone resulted in the conformers I and II, respectively. Theoretical criteria (max(Delta theta), rmsd[r]-crystal, energy profile) confirmed that the structural differences in the conformers are in the range of acceptable values for the detection of conformational changes. The phase stability of the polymorphs was investigated by slurry and grinding methods as well as by the HSM technique. Since the orientations of pyrazine and phenyl moieties were altered in the polymorphs, the hydrogen bond donor and acceptors exhibited meaningful supramolecular architectures in the crystal packing as well as C-HMIDLINE HORIZONTAL ELLIPSISN, C-HMIDLINE HORIZONTAL ELLIPSISO and C-HMIDLINE HORIZONTAL ELLIPSIS pi interactions. The energetic study of the noncovalent interactions in the molecular pairs (dimers) of the polymorph crystal structures was performed by DFT-D calculations.

A series of tripeptidic proteasome inhibitors with furylketone as C-terminus were designed and synthesized. Biochemical evaluations against beta 1, beta 2 and beta 5 subunits revealed that they acted selectively on beta 5 subunit with IC(50)s against chymotrypsin-like (CT-L) activity in micromolar range. LC-MS/MS analysis of the ligand-20S proteasome mixture showed that the most potent compound 11m (IC50 = 0.18 mu M) made no covalent modification on 20S proteasome. However, it was identified acting in a slowly reversible manner in wash-out assay and the reversibility was much lower than that of MG132, suggesting the possibility of these tripeptidic furylketones forming reversible covalent bonds with 20S proteasome. Several compounds were selected for anti-proliferative assay towards multiple cancer cell lines, and compound 11m displayed comparable potency to positive control (MG132) in all cell lines tested. Furthermore, the pharmacokinetic (PK) data in rats indicated 11m behaved similarly (C-max, 2007 mu g/L; AUC(0-t), 680 mu g/L.h; V-ss, 0.66 L/kg) to the clinical used agent carfilzomib. The yield of the pyrazine-2-carboxamides and the reaction time depended on the type of the amine (aliphatic vs aromatic), substitution pattern, and number of substituents on the aromatic amines.N-(4-chlorophenyl)pyrazine-2-carboxamides can be prepared by this method in 81% yield;N-(2-ethylhexyl)pyrazine-2-carboxamide andN-(4-fluorobenzyl)pyrazine-2-carboxamide showed the best activity againstMycobacterium tuberculosisH37Rv (<6.25 mu g/mL). Photophysical properties of these coordination polymers are studied, and they show characteristic emissions of corresponding Ln(III) ions which should be attributed to the sensitization of the Agligand section (d-block). In addition, under the co-action of crystal field and 4d orbits of Ag(I) ion, the 4f orbits of Ln(III) ions are tuned and cause obvious shift of partial energy levels, which present shifts of the corresponding NIR emission bands and can be corroborated in their UV-Vis-NIR absorption spectra. The C-scorpionate iron(II) complex [FeCl2(Tpm)] [Tpm = kappa(3)-HC(C3H3N2)(3)] was immobilized on three nanoporous carbon supports to produce active, selective and recyclable catalysts for the oxidation of cyclohexane. The influence of the porous carbon supports on the performance of the heterogenized [FeCl2(Tpm)] catalyst was investigated using materials with distinct porosity: microporous (GL50-ox, wet oxidized GL50 Norit sample, and S, sisal-derived activated carbon prepared by chemical activation), and mesoporous (CMK-3) materials. The heterogenized systems exhibited good activity and rather high selectivity to the formation of KA (ketone-alcohol) oil (cyclohexanol and cyclohexanone mixture) from microwave-assisted oxidation of cyclohexane, and allowed their easy recovery and reuse, at least for four consecutive cycles. The most important outcome of this work was the significant self-catalytic activity observed for the pristine carbon materials GL50-ox and CMK-3 towards cyclohexane oxidation under Microwave irradiation, in the absence of the iron complex.

We herein report the conventional and microscale parallel synthesis of selective inhibitors of human blood coagulation factor XIIa and thrombin exhibiting a 1,2,4-triazol-5-amine scaffold. Structural variations of this scaffold allowed identifying derivative 21i, a potent 29 nM inhibitor of FXIIa, with improved selectivity over other tested serine proteases and also finding compound 21m with 27 nM inhibitory activity toward thrombin. For the first time, acylated 1,2,4-triazol-5-amines were proved to have anticoagulant properties and the ability to affect thrombin- and cancer-cell-induced platelet aggregation. Performed mass spectrometric analysis and molecular modeling allowed us to discover previously unknown interactions between the synthesized inhibitors and the active site of FXIIa, which uncovered the mechanistic details of FXIIa inhibition. Synthesized compounds represent a promising starting point for the development of novel antithrombotic drugs or chemical tools for studying the role of FXIIa and thrombin in physiological and pathological processes.
