The tert-butyl 4-(5-cyanopyrazin-2-yl)piperazine-1-carboxylate required for the synthesis was prepared in the following manner: To a solution of 5-chloropyrazine-2-carbonitrile (200 mg, 1.43 mmol) and tert-butyl piperazine-1-carboxylate (266.95 mg, 1.43 mmol) in anhydrous DMA (5 ml) under N2 was added DIPEA (748.95 muL, 4.3 mmol). The mixture was stirred and heated at 150C in a microwave for 90 mins. The mixture was allowed to cool to RT, prior to adding water until a significant amount of precipitate was observed. This was filtered under vacuum, washed with water and dried to afford 363mg, 87% of titled compound as a light brown solid.
