The tuning effect of C3-ester groups on the glycosylation stereochemistry of L-rhamnopyranose (L-Rha) ethyl thioglycoside donors is described. On one hand, the L-Rha thioglycoside donors carrying 3-O-arylcarbonyl or levulinoyl group undergo highly alpha-selective glycosylation to afford a wide variety of alpha-L-rhamnoside products in high chemical yields. On the other hand, the glycosylation of the 3-O-4-nitropicoloyl and 2-pyrazinecarbonyl group substituted L-Rha thioglycosides displays beta-stereoselectivity. Only or predominant beta anomeric products are obtained when these L-Rha donors couple with the primary or reactive secondary acceptors, while the beta-selectivity may decrease significantly when these donors react with less reactive secondary alcohols. The synthetic utility of the newly developed alpha- and beta-directing L-Rha donors 1h and 1e has been demonstrated by the efficient synthesis of a structurally unique trisaccharide 9, which is derived from the cell wall polysaccharide of Sphaerotilus natans.

A universally applicable method for the prediction of the isobaric heat capacities of the liquid and solid phase of molecules at 298.15 K is presented, derived from their true volume. The molecules’ true volume in A(3) is calculated on the basis of their geometry-optimized structure and the Van-der-Waals radii of their constituting atoms by means of a fast numerical algorithm. Good linear correlations of the true volume of a large number of compounds encompassing all classes and sizes with their experimental liquid and solid heat capacities over a large range have been found, although noticeably distorted by intermolecular hydrogen-bond effects. To account for these effects, the total amount of 1303 compounds with known experimental liquid heat capacities has been subdivided into three subsets consisting of 1102 hydroxy-group-free compounds, 164 monoalcohols/monoacids, and 36 polyalcohols/polyacids. The standard deviations for Cp(liq,298) were 20.7 J/mol/K for the OH-free compunds, 22.91 J/mol/K for the monoalcohols/monoacids and 16.03 J/mol/K for the polyols/polyacids. Analogously, 797 compounds with known solid heat capacities have been separated into a subset of 555 OH-free compounds, 123 monoalcohols/monoacids and 119 polyols/polyacids. The standard deviations for Cp(sol,298) were calculated to 23.14 J/mol/K for the first, 21.62 J/mol/K for the second, and 19.75 J/mol/K for the last subset. A discussion of structural and intermolecular effects influencing the heat capacities as well as of some special classes, in particular hydrocarbons, ionic liquids, siloxanes and metallocenes, has been given. In addition, the present method has successfully been extended to enable the prediction of the temperature dependence of the solid and liquid heat capacities in the range between 250 and 350 K.

Treatments of N-(pyridin-2-ylmethyl)pyrazine-2-carboxamide (L-1), N-(quinolin-8-yl)pyrazine-2-carboxamide (L-2), N-(quinolin-8-yl)picolinamide (L-3) and N-(quinolin-8-yl)quinoline-2-carboxamide (L-4) with [PdCl2(NCMe)](2) afforded the corresponding Pd(ii) complexes, [Pd(L-1)Cl] (PdL1); [Pd(L-2)Cl] (PdL2); [Pd(L-3)Cl] (PdL3); and [Pd(L-4)Cl] (PdL4) in moderate yields. Structural characterisation of the compounds was achieved by NMR and FT-IR spectroscopies, elemental analyses and single crystal X-ray crystallography. The solid-state structures of complexes PdL2-PdL4 established the presence of one tridentate carboxamide and Cl ligands around the Pd(ii) coordination sphere, to give distorted square planar complexes. Electrochemical investigations of PdL1-PdL4 showed irreversible one-electron oxidation reactions. Kinetics reactivity of the complexes towards bio-molecules, thiourea (Tu), l-methionine (L-Met) and guanosine 5 ‘-diphosphate disodium salt (5 ‘-GMP) decreased in the order: PdL1 > PdL2 > PdL3 > PdL4, in tandem with the density functional theory (DFT) data. The complexes bind favourably to calf thymus (CT-DNA), and bovine serum albumin (BSA), and the order of their interactions agrees with the substitution kinetics trends. The in vitro cytotoxic activities of PdL1-PdL4 were examined in cancer cell lines A549, PC-3, HT-29, Caco-2, and HeLa, and a normal cell line, KMST-6. Overall, PdL1 and PdL3 displayed potent cytotoxic effects on A549, PC-3 HT-29 and Caco-2 comparable to cisplatin. All the investigated complexes exhibited lower toxicity on normal cells than cisplatin.

The Yamaguchi reaction is widely and generally applied to synthesize esters and lactones. It involves 2,4,6-trichlorobenzoyl chloride as the Yamaguchi reagent, 4-dimethylaminopyridine, and triethylamine. Pyrazinamide is a crucial first-line drug for tuberculosis treatment, therefore their analogues are interesting in organic synthesis. In general, the synthesis pyrazinamide analogues involve reaction of pyrazine-2-carboxylic acids with thionyl chloride to yield the corresponding acyl chlorides, which on treatment with amines gave pyrazine-2-carboxamides. However, thionyl chloride is listed under the Chemical Weapons Convention and releases toxic sulfur oxide when react with carboxylic acid. We successfully synthesized series of pyrazinamide analogues using the Yamaguchi reaction. The synthesis involved reaction of pyrazine-2-carboxylic acid with various aliphatic and aromatic amines in the presence of Yamaguchi reagent and 4-dimethylaminopyridine. The yield of the pyrazine-2-carboxamides and the reaction time depended on the type of the amine (aliphatic vs aromatic), substitution pattern, and number of substituents on the aromatic amines.N-(4-chlorophenyl)pyrazine-2-carboxamides can be prepared by this method in 81% yield;N-(2-ethylhexyl)pyrazine-2-carboxamide andN-(4-fluorobenzyl)pyrazine-2-carboxamide showed the best activity againstMycobacterium tuberculosisH37Rv (<6.25 mu g/mL). This result could lead to find more active pyrazinamide analogues. A set of new salts containing benzimidazole and pyrazine groups, namely, [HPZBI]X-+(-) nH(2)O (X = Cl (n = 1) (1), NO3 (n = 1) (2), ClO4 (n = 0) (3) and H2PO4 (n = 0) (4)) [PZBI = 2-(pyrazin-2-yl)-1H-benzimidazole], were obtained by the reaction of PZBI and a set of inorganic acid, respectively. The emission maxima can be found at 444, 472, 471, 432 and 443 nm for PZBI and its compounds 1-4 in the solid state at room temperature, respectively. Compared with the free PZBI, the emission maxima of compounds 1-4 are obviously blue/red-shifted for 1-4, indicating that the emission maxima are relative to the intermolecular packing distances. Their photoluminescent lifetime and quantum yield are 1.10 ns and 24.5% for 1, 0.99 ns and 26.4% for 2, 0.94 ns and 21.65% for 3, 2.86 ns and 20.57% for 4, respectively. Compounds 1-4 were structurally characterized by X-ray single crystal diffraction, FT-IR, and UV-Vis spectra. Single crystal X-ray diffraction reveals that pi center dot center dot center dot pi packing interactions and a set of hydrogen bonds can be observed. The shortest distances of pi center dot center dot center dot pi stacking interactions are 3.613, 3.534, 3.731 and 3.492 angstrom in compounds 1-4, respectively. Additionally, the thermal stabilities of compounds 1-4 were investigated in detail.

A high-nuclearity nickel-substituted polyoxotungstate, Na6K5.5H4.5{[Ni-8(OH)(6)(H2O)(2)](CO3)(3)(SiW9O34)(2)}center dot 22H(2)O (1), has been obtained by the reaction of Na-10[alpha-SiW9O34]center dot 18H(2)O, Ni(NO3)(2)center dot 6H(2)O and K(2)CO(3)in aqueous solution and characterized by IR spectroscopy, single crystal X-ray diffraction, thermal gravimetric analysis and elemental analysis. Carbonate is not only used to adjust the pH of the solution, but also is a structure-stabilizing agent. The polyoxoanion {[Ni-8(OH)(6)(H2O)(2)](CO3)(3)(SiW9O34)(2)}(16-)(1a) can be regarded as two {[Ni-4(OH)(3)(H2O)](SiW9O34)} subunits connected by three carbonate groups, which represents the first polyanion containing an octanuclear nickel cluster based on trivacant Keggin-type polyoxotungstate {XW9} (X = heteroatom). The subunit {[Ni-4(OH)(3)(H2O)](SiW9O34)} can be viewed as a tetranuclear nickel cluster linked to a trivacant Keggin-type polyoxotungstate {SiW9O34}. Candida albicans biofilm represents a major clinical problem due to its intrinsic tolerance to anti-fungal compounds and it has been highly related to infections in catheterized patients. Few compounds are described as able to inhibit biofilm formation or to interfere with preformed biofilm of C. albicans. Here we report the in vitro evaluation of anti-biofilm activity on C albicans ATCC 10231 of a series of new and already known amine and amide indole derivatives. Among the studied compounds, fifteen resulted active on C albicans ATCC 10231 biofilm, with BMIC50 <= 16 mu g/mL. Three of them (7, 23 and 33) showed a selectivity towards mature biofilm and the most active of them was the compound 23 (BMIC50 = 4 mu g/mL). On the other hands, two different compounds (21 and 22) were selective towards biofilm formation with BMIC50 values of 8 mu g/mL Otherwise, compounds 16 and 17 resulted active on biofilm formation, with BMIC50 of 8 mu g/mL and 2 mu g/mL respectively, and on mature biofilm with BMIC50 of 2 mu g/mL. These two last compounds also showed an interesting activity towards the planktonic cells of C albicans. A selection of the more active compounds was also evaluated on different C albicans strains (PMC1042, PMC1083 and ATCC 10261), showing a comparable or higher anti-biofilm activity, especially on mature biofilm. In vivo toxicity studies using the Galleria mellonella larvae, were finally carried out on more active indole derivatives, showing that they are poorly toxic even at the highest concentrations tested (500-1000 mu g/mL). Although pyrazinamide (PZA) is a key component of first- and second-line tuberculosis treatment regimens, there is no gold standard to determine PZA resistance. Approximately 50% of multidrug-resistant tuberculosis (MDR-TB) and over 90% of extensively drug-resistant tuberculosis (XDR-TB) strains are also PZA resistant. pncA sequencing is the endorsed test to evaluate PZA susceptibility. However, molecular methods have limitations for their wide application. In this study, we standardized and evaluated a new method, MODS-Wayne, to determine PZA resistance. MODS-Wayne is based on the detection of pyrazinoic acid, the hydrolysis product of PZA, directly in the supernatant of sputum cultures by detecting a color change following the addition of 10% ferrous ammonium sulfate. Using a PZA concentration of 800 mu g/ml, sensitivity and specificity were evaluated at three different periods of incubation (reading 1, reading 2, and reading 3) using a composite reference standard (MGIT-PZA, pncA sequencing, and the classic Wayne test). MODS-Wayne was able to detect PZA resistance, with a sensitivity and specificity of 92.7% and 99.3%, respectively, at reading 3. MODS-Wayne had an agreement of 93.8% and a kappa index of 0.79 compared to the classic Wayne test, an agreement of 95.3% and kappa index of 0.86 compared to MGIT-PZA, and an agreement of 96.9% and kappa index of 0.90 compared to pncA sequencing. In conclusion, MODS-Wayne is a simple, fast, accurate, and inexpensive approach to detect PZA resistance, making this an attractive assay especially for low-resource countries, where TB is a major public health problem.
