A common strategy employed in antibacterial drug discovery is the targeting of biosynthetic processes that are essential and specific for the pathogen. Specificity in particular avoids undesirable interactions with potential enzymatic counterparts in the human host, and it ensures ontarget toxicity. Synthesis of pantothenate (Vitamine B5), which is a precursor of the acyl carrier coenzyme A, is an example of such a pathway. In Mycobacterium tuberculosis (Mtb), which is the causative agent of tuberculosis (TB), pantothenate is formed by pantothenate synthase, utilizing D-pantoate and beta-Ala as substrates. beta-Ala is mainly formed by the decarboxylation of L-aspartate, generated by the decarboxylase PanD, which is a homo-oliogomer in solution. Pyrazinoic acid (POA), which is the bioactive form of the TB prodrug pyrazinamide, binds and inhibits PanD activity weakly. Here, we generated a library of recombinant Mtb PanD mutants based on structural information and PZA/POA resistance mutants. Alterations in oligomer formation, enzyme activity, and/or POA binding were observed in respective mutants, providing insights into essential amino acids for Mtb PanD’s proper structural assembly, decarboxylation activity and drug interaction. This information provided the platform for the design of novel POA analogues with modifications at position 3 of the pyrazine ring. Analogue 2, which incorporates a bulky naphthamido group at this position, displayed a 1000-fold increase in enzyme inhibition, compared to POA, along with moderately improved antimycobacterial activity. The data demonstrate that an improved understanding of mechanistic and enzymatic features of key metabolic enzymes can stimulate design of more-potent PanD inhibitors.

We utilized synthetic photochemistry to generate novel sp 3 -rich scaffolds and report the design, synthesis, and biological testing of a diverse series of amides based on the 1-(amino-methyl)-2-benzyl-2-azabicyclo[2.1.1]hexane scaffold. Preliminary antimalarial screening of the library provided promising compounds with activity in the 1-5 mu M range with an enhanced hit rate. A new chromone based Schiff base ligand L was synthesized by the condensation of 3-formyl chromone and pyrazine-2-carbohydrazide as a colorimetric probe to detect Cu (II) ions selectively. An instant visual colour change from colourless to yellow was obtained on addition of Cu2+ ions to the probe L solution, while other metal ions found ineffective. The ligand L was characterized by H-1 NMR, FTIR and HRMS spectral techniques. UV-Visible spectroscopic technique was used to study the sensing ability of probe L for copper ions above other metal ions. The Job’s plot obtained from absorption studies and HRMS data confirmed that the Cu2+ ions bind with ligand L in 1:1 stoichiometric ratio. DFT computations were also supported the binding framework between L and Cu (II) ions. The LOD value and the association constant were obtained 3.9 x 10(-7) M and 2.3 x 10(5) M-1 respectively, via Benesi-Hildebrand equation. Selectivity of L towards Cu2+ ions was also studied and it was found that the probe L worked specifically for copper ions without any considerable influence of other intruding metal ions. Based on the interaction modes of the natural 20S proteasome inhibitors TMC-95A, we have previously discovered a dipeptide 1. To explore the SAR around compound 1, we designed and synthesized a series of dipeptides (8-38) with a fragment-based strategy. Among them, nine compounds showed significant inhibitory activities against the chymotrypsin-like activity of human 20S proteasome with IC50 values at the submicromolar level, which were comparable or even superior to the parent compound 1. Meanwhile, they displayed no significant inhibition against trypsin-like and caspase-like activities of 20S proteasome. The results suggested the feasibility to design dipeptides as novel and potent 20S proteasome inhibitors.

The synthesis of modified RGD peptides and their conjugation to the pyrazine skeleton at their N-terminus are described. To modify and alter the RGD sequence, short bioactive peptides such as FALKF and NGRG were added to the RGD N-terminus. Moreover, the in vitro investigation of these modified peptides, by a cell adhesion assay using the melanoma cell lines M21 expressing alpha(v)beta(3) and M21L lacking alpha(v) expression, was made and interestingly peptide 4 containing the pyrazine moiety with the linear RGDFAKLF sequence gave the best IC50 value with M21 and all the peptides were unable to bind to M21L, demonstrating the selective recognition to alpha(v)beta(3). The results showed pyrazine plays an essential role in the activity of the peptides. From these results, we can suggest that these peptides can affect cancer cells by abrogation of cell adhesion (cell migration).

A series of 7-benzamidocoumarin derivatives 10-25 starting from 7-amino coumarins 7 and 8 has been synthesized, characterized and evaluated in vitro for antifilarial activity against the human lymphatic filarial parasite, Brugia malayi. Compounds 13 and 20-23 showed permanent paralysis of the worm with 90-95% inhibition of motility of adult worms at 10 mu M. All the synthesized compounds were docked on the modeled receptor of Onchocerca volvulus chitinase OvCHT1. Compound 13 with binding energy of -7.95 Kcal/mol showing three hydrogen bonds with the active site of the enzyme emerged as the best inhibitor.

Complexation of U(vi) with pyridazine-3-carboxylate (PDZ) and pyrazine-2-carboxylate (PAZ) was studied by spectrophotometry, potentiometry and microcalorimetry in 1.0 mol dm(-3) NaClO4. Three complexes, [UO2L](+), UO2L2(aq) and [UO2L3](-), were identified and their stability constants (log) and the corresponding formation enthalpies were determined. The thermodynamic parameters indicate that the formation of the three complexes is endothermic and driven exclusively by entropy. H-1 and C-13-NMR data provide insight into the coordination modes of the complexes which corroborate with the thermodynamic data. Ligands chelate to U(vi) via (2)(N,O) coordination mode in complexes [UO2L](+) and UO2L2(aq). The crystal structures of four U(vi) complexes, [(UO2)(PAZ)(2)(H2O)]H2O(i), [(UO2)(PDZ)(2)(H2O)](ii), [(UO2)(PDZ)(3)Na2ClO4]2H(2)O(iii), and [(UO2)(2)(PDZ)(4)(H2O)(2)]2H(2)O(iv), were determined by single-crystal X-ray diffraction and compared with the U(vi) complex with picolinate (PA) (CH6N3)[UO2(PA)(3)] in the literature. The structure data suggest that the carboxylates coordinate with uranium in O?C-O-U mode. The strengths of the U-O-C-C-N chelate cycles in the U(vi)/L complexes decrease with the trend of PA > PDZ > PAZ, which is in great agreement with the trend of thermodynamic parameters in aqueous solutions. It is interesting that in compound II two PDZ molecules coordinate with U(vi) in cis-planar positions via (2)(N,O) mode, but in other metal complexes of the three ligands having the same (2)(N,O) coordination mode the two ligand molecules are all in trans-arrangement. In the dimeric complex IV, one ligand coordinate with U(vi) in (2)(N,O) mode, while the other does it in (2)-L-(2)(O:O) mode respectively.

Mitochondrial complex III is one of the most promising targets for a number of pharmaceuticals and fungicides. Due to the wide-spread use of complex III-inhibiting fungicides, a considerable increase of resistance has occurred worldwide. Therefore, inhibitors with novel scaffolds and potent activity against complex III are still in great demand. In this article, a new series of amide compounds bearing the diaryl ether scaffold were designed and prepared, followed by the biological evaluation. Gratifyingly, several compounds demonstrated potent activity against succinate-cytochrome c reductase (SCR, a mixture of mitochondrial complex II and complex III), with compound 3w possessing the best inhibitory activity (IC50 = 0.91 +/- 0.09 mu mol/L). Additional studies verified that 3w was a new inhibitor of complex III. Moreover, computational simulations elucidated that 3w should bind to the Q(o) site of complex III. We believe this work will be valuable for the preparation and discovery of more complex III inhibitors.

A novel and regioselective protocol for C-S coupling of naphthylamines via the insertion of sulfur dioxide was developed by employing a heterogeneous copper catalyst, providing desired products in moderate to good yields. This strategy gives a powerful tool for the efficient preparation of sulfur-containing compounds. Control experiments declared that a single-electron transfer mechanism (SET) is responsible for this C-S cross coupling reaction.
